The study of the neuroprotective effect of the extract of the insect in vitro

Insects and the substances extracted from them have been used as medicinal resources by human cultures all over the world. The substances extracted from darkling beetles ( Blaps femoralis ) has been used as antimicrobial, analgesic, anti ulcer agents in Asian countries. Aim of this study was to evaluate neuroprotective effects against neurotoxicity in vitro and clarify possible mechanism. Results: Blaps femoralis extract has been demonstrated to scavenge hydroxyl radicals by ESR. Half-maximal inhibitory concentration, IC50 of the extract was 11.35 mg/ml. It restored the decreased level of complex-I, MDH activities in a concentration of 8.0, 17.0 and 35.0 mg/ml. The neuroprotective effects were assessed by HO-induced neurotoxicity in primary cultured cortical neurons. Also, pretreatment with the extract Blaps femoralis (0.5 mg/ml) was found to impart significant protection against HO-induced neurotoxicity in primary cultured cortical neurons. Furthermore, the use of a luciferase reporter (ARE-lic) assay showed that extract from Blaps femoralis activates protective signaling pathway in primary cortical neurons through ARE/Nrf2 pathway.
