The volume of the frozen sample as one of factors of survival of spermatozoa of sturgeon species at the cryopreservation

The sexual products of the Russian sturgeon (Acipenser gueldenstaedtii Brandt, 1833), the Siberian sturgeon of Lena’s population (Acipenserbaerii Brandt, 1869) were used as objects of research. The purpose of represented work was studying of dependence of volume of the container in which freezing of sperm liquid and survival the defrosting spermatozoa of sturgeon species was carried out. The greatest time of life of spermatozoa, at realization of two series of experiences with different types of sturgeon fishes, the samples, frozen in test tubes of Eppendorf of 0,5 ml, was observed. Cryopreservation of sperm liquid in test tubes of bigger volume (0.75 ml; 1.50 ml and 2.00 ml) showed results slightly worse. Direct dependence between the volume of a frozen material and survival of defrosting sperm was established. With increasing of the volume of a frozen sample safety of the frozen-thawed cells falls as there is a dispersion of speed of cooling in a frozen sample at which the most part of cells is frozen with a speed different from optimum value. The received results allow drawing a conclusion on primary use of smaller volume of capacity at low-temperature conservation. It will promote increase of efficiency of developed techniques of a cryopreservation of a genetic material.
