Influence of aromatic sulphonamide derivatives, potential inhibitors of carboxyanhydrase 9 on viability and intracellular pH of MDCK (Madine-Darby Canine Kidney cells)

Carboxyanhydrase 9 is a protein, which recently was reported to be promising oncogene marker. This protein helps tumor cells keep intracellular pH within physiologically grade and acidify extracellular pH instead. It was supposed that inhibition of carboxyanhydrase 9 can lead to suppression of hypoxic tumors. That’s why inhibitors of carboxyanhydrase 9 were proposed to be promising antitumor agents. MDCK (epithelial cells of the canine kidney) were treated by 6 newly synthesized aromatic sulphonamides, as it is known that this chemical group selectively binds to carboxyanhydrase 9. Before experiments, MDCK cells were transfected in the way, that one of cell lines constantly produces carboxyanhydrase 9 and other is a negative control. Potential inhibitors were incubated with cells in the atmosphere with normal concentration and with lack of oxygen. Finally it was shown, that the chemicals are reducing cell viability after 24 and 48 hours and no decrease of intracellular pH of cells were observed comparing to control experiments. So, we conclude, that the inspected substances do not specifically bind to carboxyanhydrase 9. At the same time it was shown that the developed technique with use of SNARF can be applied for measurement of intracellular pH for MDCK cells. Also, it was shown that hypoxic regime of cultivation leads to acidification of intracellular pH, which is in accordance with previously reported data.
