Cytological description of the cell culture MDCK-II (Madine-Darby Canine Kidney cells) during different stages of apoptosis by means of biosensor based on quartz crystal microbalance

Apoptosis is a complex biochemical process, which is initiated in living cells by different stimuli. New biosensor system was applied for monitoring of the apoptosis in MDCK-II (epithelial cells of the canine kidney). Cells were cultivated in cellular incubator in Petri dishes and in the microreactor of biosensor system on the surface of quartz crystal microbalance. Previously it was shown, that apoptosis is responsible for great changes in cytoskeleton. That was the reason why it was decided to use quarts crystal microbalance to detect cytoskeleton rearrangements on different stages of apoptosis. Cells were treated with different concentrations of bleomycin, which is a chemical substance, widely used for initiation of apoptosis in cell cultures and in vivo. Also, bleomycin is known for it’s antitumor action. The cytological description of cell culture on the different stages of apoptosis was presented as a result of research. The description is based on light microscopy data, fluorescent microscopy data and on changes in signal from impedance analyzer, connected to quartz crystal microbalance. After several hours cells being incubated with bleomycin, amount of necrotic cells raised under both regimes of cultivation. It was shown, that the process of apoptosis in the cells under microfluidic flow is initiated in less crucial manner, than in standard cell culture.
